Dier went off against Burnley with an apparent calf problem but is able to play against the Saints.
The hosts make two changes, with Mousa Dembele and Danny Rose coming in for Ben Davies and Moussa Sissoko.
A goal for Harry Kane would see the Spurs striker set a new Premier League scoring record for a single year.
Meanwhile, Southampton are without the injured Cedric Soares and Ryan Bertrand as well as the suspended Charlie Austin.
Former QPR striker Austin is replaced by Shane Long.
